abstractText  Full text of Mouse Genome contribution: Research News: 1. Hesitant (hes). Hesitant (hes), a new recessive neurological mutation, appeared in the C3H/HeJ-ca (acromelanic) strain maintained in the Mouse Mutant Resource of The Jackson Laboratory in 1987. Ovaries from an affected female were transplanted to rescue the mutation. Hesitant is now maintained on the C3HeB/FeJLe background by brother x sister matings. Homozygotes of both sexes are fertile, although fertility in males on this background seems to be reduced. Homozygotes, hes/hes, can be recognized as early as 14 days of age by their slightly smaller size and hesitant movement when attempting to walk. Identification of hes mutants is easier at 21 days of age. Affected mice start to move forward by lifting one hind foot then the other into the air. After a slight hesitation, the feet are placed flat on the surface with the rear legs extended stiffly causing the posterior to rise, and the mouse moves forward. Front feet and legs are not affected. Entire body tremors are not present. No spasticity nor muscle degeneration has been observed. The abnormal gait persists throughout the lifetime of the mutant. Hes has been located on Chr 10 linked to steel. Backcrosses of Sl +/+ hes females x + hes/+ hes males have produced 33 Sl +, 34 + hes, 12 + +, 3 Sl hes (N=82). Backcrosses of Sl +/+ hes males x + hes/+ hes females have produced 40 Sl +, 28 + hes, 3 + +, 5 Sl hes (N=76). Combining the results of the two crosses gives a recombination estimate of 14.56 +/- 2.8% between Sl and hes. Further crosses to determine gene order on Chr 10 are in progress. (Sweet)
