| molecularNote | A loxP-flanked neomycin resistance cassette replaced exon D, which encodes sequences for the beta1D alternative isoform. The neomycin cassette was deleted by Cre-mediated recombination in ES cells prior to blastocyst injection, leaving a single loxP site instead of exon D. Northern blot analysis revealed that no beta1D specific transcript was present, but transcripts of other splice variants of the gene were not afftected by this mutation. Western blot analysis confirmed the absence of the beta1D specific protein. |
