| molecularNote | 2.2 kb of the gene, which contains the first 3 exons encoding the translation start site and most of the homeodomain, was replaced with a PGK-neo cassette via homologous recombination. RNase protection assay showed absence of gene expression in placentas and urogenital ridges of E12.5 hemizygous male and homozygous female mutant embryos. A small band intermittently observed may represent the alternative expression of 151 nt of exon 4. RT-PCR using primers to exons 2 and 4 further demonstrated the absence of gene transcription in the urogenital ridge of E12 mutant embryos. |
