| molecularNote | A duplication of exon 6 (an exon common to all known splice variants) was inserted into intron 6 along with stop codons, a polyadenylation signal, and a neo gene. A frameshift arises from the splicing of exon 6 to its duplicate, resulting in the substitution of the isoform-specific carboxy terminal sequences with a new carboxy terminal sequence encoding 11 residues. RT-PCR, immunoprecipitation, and immunolabeling analyses showed that the mutant isoform was the only isoform produced and that the altered carboxy terminal region did not affect localization of the protein in homozygous mutant mice. |
