| molecularNote | The Dad1 locus was disrupted by the insertion of a neomycin selection cassette that replaced non-coding exon 3 of Dad1 and the Tcra associated LCR (DNAse I hypersensitive sites 2 through 6; Rr312). Neither Dad1 transcript nor protein was detected in homozygous mutant mice by RNAse protection or Western blot analyses. Expression analysis showed the deleted hypersensitive sites were necessary for the enhancement of Tcra transcription in a fraction of immature T cells. |
