| molecularNote | Two loxP sites were introduced to intronic regions flanking the three exons encoding part of the lumenal domain, the transmembrane domain, and part of the catalytic domain. A third loxP site was introduced with a PGK-neo cassette directly downstream of the second loxP site in the same intron. The three exons and PGK-neo cassette were excised by mating chimeric animals to a cre-expressing line. RT-PCR analysis of homozygous mutant animals using primers flanking the deleted region confirmed that gene transcripts did not contain the deleted exons. Western blot analysis using antibodies directed against the kinase domain did not detect protein product in pancreatic lysates from homozygous mutants. |
