| molecularNote | A floxed neo cassette replaced 3.7 kb of sequence encompassing a CpG island, transcriptional start sites, and the putative promoter. Absence of a 129-specific MnlI restriction pattern indicated that the 129-derived X chromosome was targeted. Expression analysis showed a significant reduction in, if not a complete ablation of, transcription from the targeted allele, though transcript was shown to be produced from the normal X on the untargeted chromosome. Residual transcription was only detected by increased PCR cycles, though it was not clear if this weak transcription was due to minor and/or cryptic promoters or rather to overcylcing conditions. No residual transcription was detected by RNA FISH. |
