| molecularNote | Exons 1 and 2 were replaced with an IRES-lacZ-floxed-neo cassette via homologous recombination. The floxed neo cassette was removed in correctly targeted ES cells by transient expression of cre recombinase. Absence of gene expression in homozygous mutant animals was demonstrated by Western blot of hindbrain and forebrain extracts using anti-POMA antisera which recognizes all species of nova proteins. The Nova1-specific band was not observed in samples from homozygous mutants. |
