| molecularNote | A targeting vector containing a Tcrg-V2 gene modified to lack a restriction site, a floxed Tcrg-V4 gene, and a Tcrg-V3 gene also modified to lack a restriction site was electroporated into ES cells. The ES cells were then transfected with cre recombinase, resulting in the excision of Tcrg-V4. Tcrg-V2 was left adjacent to Tcrg-V3 (with single loxP site in between). Southern blot analysis indicated that only a portion of the targeting vector recombined as the modified Tcrg-V2 and the endogenous Tcrg-V3 (not the modified Tcrg-V3) were present. |
