| molecularNote | The endogenous locus was disrupted by the insertion of a neomycin cassette that replaced sequence extending from 252 bp upstream of the TATA box through exon 2. While neither normal transcript or protein was detected by Northern or Western blot analyses of homozygous mutant mice, an elongated transcript and a 28 kDa protein were observed. Analysis of labeled retinol in serum fractions and in vivo binding experiments indicated that the mutant protein was present in high molecular weight aggregates and did not function to transport retinol in circulation. |
