| molecularNote | The transgene consists of a mouse Cd4 promoter and a flag-tagged mutant human CUL1 sequence encoding 252 N-terminal residues. Mouse Cd4 regulatory sequences, including the minimal enhancer, minimal promoter, 70 bp of exon 1, and part of intron 1, were used to drive transgene expression in T lymphocytes. The Cd4 promoter lacks the silencer region and is transcriptionally active in both single- and double-positive T cells. Transgene expression was confirmed by Western blot analysis of thymocyte extracts from transgenic animals. The transgenic human protein was shown to associate with mouse Skp1 protein via immunoprecipitation immunoblot. |
