Allele : Tg(Cd4-CUL1-N252)1Ingh transgene insertion 1, Giorgio Inghirami

Primary Identifier  MGI:2447878 Allele Type  Transgenic
Attribute String  Inserted expressed sequence Gene  Tg(Cd4-CUL1-N252)1Ingh
Strain of Origin  Not Specified Is Recombinase  false
Is Wild Type  false
description  Three transgenic lines (4, 10, and 20) were created. Highest expressing lines 10 and 20 were used for analysis.
molecularNote  The transgene consists of a mouse Cd4 promoter and a flag-tagged mutant human CUL1 sequence encoding 252 N-terminal residues. Mouse Cd4 regulatory sequences, including the minimal enhancer, minimal promoter, 70 bp of exon 1, and part of intron 1, were used to drive transgene expression in T lymphocytes. The Cd4 promoter lacks the silencer region and is transcriptionally active in both single- and double-positive T cells. Transgene expression was confirmed by Western blot analysis of thymocyte extracts from transgenic animals. The transgenic human protein was shown to associate with mouse Skp1 protein via immunoprecipitation immunoblot.
