| molecularNote | The nLacZ gene and a loxP site flanked neomycin resistance gene cassette were inserted in place of codons 1-125, which encode the signal sequence and most of the cystein-rich ligand-binding domain. PCR confirmed integration of neo and the absence of endogenous sequence and X-gal staining of mutant skin showed that the gene was expressed. Western blot against the C-terminal 17 amino acids of the endogenous gene demonstrated its absence in mutant skin. The floxed neo was later removed through crossing with germ-line Cre-expressing mice. |
