molecularNote  A targeting vector containing exon 3 (the coding exon) of Gjc1 (Gja7), a floxed neo, and exon 2 (the coding exon) of Gja5 with a loxP site near the 3' end was inserted immediately after the start site of the endogenous gene, within exon 2, so as to allow for the expression of Gjc1 in place of Gja5. The second amino acid of Gjc1 was mutated from a serine to a glycine. Mice transmitting the mutation through the germ line were crossed with either CAG-Cre or Cre-deleter mice to excise the floxed neo and most of exon 2 of Gja5. Southern blot and PCR confirmed recombination. RT-PCR, Northern blot and Western blot indicated exon 3 from Gjc1 was expressed in knock-in mice, while Gja5 was not expressed.
