description  When mice bearing this transgene are crossed with mice transgenic for cre, excision of the loxP-flanked neo gene in cells expressing Cre recombinase results in expression of a puromycin acetyltransferase-thymidine kinase fusion protein under control of the (ubiquitously-expressing) Pgk1 promoter. Treatment of the mice with ganciclovir (GCV) causes specific ablation of cells expressing HSV1 Tk activity.
molecularNote  The coding sequence of the transgene consists of the puromycin acetyltransferase coding sequence fused in-frame to a 5' truncated HSV1 thymidine kinase gene that begins at the second ATG of the full-length gene and encodes a protein lacking the carboxy-terminal 45 amino acids. This construct resides upstream of the bovine growth hormone polyadenylation signal and downstream of the phosphoglycerate kinase 1 promoter followed by a loxP-flanked neomycin acetyltransferase gene with the bovine growth hormone poly-A signal. The transgene was inserted in reverse orientation into exon 3 of the endogenous Hprt gene via homologous recombination.
