| molecularNote | A targeting vector was designed to insert loxP sites into the start codon in exon 2 and in intron 2. A puromycin resistance cassette, flanked by FRT sites was coupled to an nlsLacZ gene in intron 2. Transient expression of Flp recombinase resulted in removal of the puro cassette. Upon cre mediated excision, the 5' end of exon 2 is removed as well as intron 2 sequence upstream of the nlslacZ. This allows the reporter to be expressed under endogenous control. |
