| molecularNote | A recombined Tcrb gene was cloned from the CD8-positive T cell clone P14, which is specific for lymphocytic choriomeningitis virus. The gene contained Vbeta8.1, Dbeta, Jbeta2.4 and Cbeta2 gene segments and the corresponding cDNA was put downstream of the H-2K promoter. Downstream of the Tcrb coding sequence, a genomic fragment of the human beta globulin gene and an IgH enhancer were added to introduce splicing sites, polyA sequence and to enhance expression. Of the four founding strains (125, 126, 128, 132) line 128 demonstrated the highest expression by flow cytometry analysis and was chosen for further study. |
