| molecularNote | A loxP site was inserted about 1kb upstream of exon 1 and a loxP-flanked neo selection cassette was inserted into intron 1. Transient transfection of Cre-recombinase deleted the genomic sequence encoding part of the promoter, the upstream untranslated region of the gene, exon 1 which contains the ATG site and part of the coding sequence for the signal peptide, and a portion of intron 1. A single loxP site remained in place of the deleted gene sequence. Gene inactivation was confirmed by Northern and Western analysis of small intestine tissue. |
