| molecularNote | This transgene comprises a 255 kb human BAC (CTD library #447M11) containing the ABCA1 gene, including 70 kb of genomic sequence upstream of the first exon and ~35 kb downstream of the 5' untranslated region. The downstream region contains one known gene, NIPSNAP3B, in opposite orientation to ABCA1. RT-PCR analysis of total RNA from various tissues demonstrates expression of human ABCA1 at levels similar to those of the endogenous mouse gene in the adrenals, brain, heart, kidney and liver and at lower levels in the intestines of transgenic mice. While basal and acetylated LDL-induced ABCA1/Abca1 expression in peritoneal macrophages is similar for human and endogenous mouse transcripts, cAMP treatment strongly induces expression of the endogenous gene, but downregulates that of the human transgene. |
