molecularNote | This transgene contains a 1.9 kb cDNA encoding a catalytically inactive mutant chicken protein tyrosine phosphatase, non-receptor type 11 (also known as SHP-2) in which the cysteine at amino acid position 459 is replaced by serine. The cDNA resides downstream of a 5.8 kb human dopamine beta-hydroxylase (dopamine beta-monooxygenase) promoter followed by the first intron of the rat insulin II gene and is joined at its 3' end to a 799 bp fragment of the human growth hormone gene that contains 66 bp of coding sequence and the 633 bp 3' untranslated region (UTR), including the polyadenylation signal. Immunoblot analysis reveals that superior cervical ganglia of transgenic mice contain 2-2.5 fold the level of DBH as do those of non-transgenic animals. |
