molecularNote | The bacterial artificial chromosome (BAC) RP24-179E13 containing the entire dopamine D1 receptor (Drd1a) gene (and incomplete Sfxn1 gene) was altered to insert a cre/ERT2 fusion gene, replacing the ATG of Drd1a ATG with the cre/ERT2 replace. This modified BAC was microinjected into the pronucleus of C57BL/6N oocytes. D1Cre-ERT2 founder mice (line 61.3) were established and found to have approximately 4 copies of the transgene. Cre recombinase expression is observed in the major projections areas of the dopaminergic system (including nucleus accumbens and striatum), while weaker expression is observed in specific regions of the cortex and thalamus, and in scattered regions of the hippocampus. |
