abstractText | The retinal neovascularization 8 mutation (rnv8) was discovered in STOCK Grhl3<ct>/J (JAX Stock No. 001813). We mated the mutant mice with C57BL/6J and all F1 mice had normal retinas. The retinal neovascularization 8 phenotype was recovered in the F2 generation at about 3 weeks of age with scattered depigmented spots of the retina, which developed into large depigmented areas by 16 weeks of age and newly formed vessels were found in the retina at both time points. Choroidal neovascularization with leakage was found by fluorescein angiography (data not show). We developed a C57BL/6J (B6) congenic strain, B6.Cg-rnv8/Boc (JAX Stock No. 008806), by backcrossing the rnv8 allele onto C57BL/6J with 4 more cycles of backcross-intercross breeding, during which Grhl3<ct> was bred out of the line, then sibling mating to homozygosity. Mice homozygous for rnv8 showed a very good rnv8 phenotype (Figure 1A) at 4 weeks of age compared with the C57BL/6J control (Figure 1B). |
