| molecularNote | A gene trap vector (pGT0Lxf) containing a splice-acceptor sequence upstream of a beta-geo reporter gene and a polyA tail was inserted into intron 2 between nucleotides 64,308,443 and 64,316,218, resulting in deletion of the corresponding part of the gene including exon 3 (nt 64,312,349-64,312,475). Semi-quantitative RT-PCR and Western blot analysis confirmed the absence of mRNA and protein expression in E11.5 homozygous mutant embryos, while X-Gal staining revealed that expression of lacZ is detectable in E10.5 heterozygous and homozygous mutant embryos. All 31 exons 3' to the insertion site of the vector are not expressed. |
