| molecularNote | A targeting cassette consisting of an EGFP downstream of an internal ribosome entry site and a floxed neomycin resistance gene replaced positions 3094-3611 in the open reading frame of the gene. The neomycin cassette was subsequently removed by crossing chimera mice with transgenic mice expressing cre recombinase in the germline. Gene inactivation was confirmed by flow cytometry analysis of activated, homozygote B cells. Low levels of GFP expression were detected in activated B cells but not in other activated lymphocytes suggesting reporter gene expression does not recapitulate expression of the endogenous locus. |
