molecularNote  The aim was to generate a novel mouse strain with a complete deletion of the p190RhoGAP-encoding gene. 1.5 kb of sequence, including the start codon and encoding the GTPase domain of exon 1, was replaced by a promoterless selection-reporter cassette encoding a Beta-Galactosidase-Neomycin resistance fusion protein in the sense orientation. The insertion site of the targeting vector is between a BamHI restriction site upstream of exon 1 and an XbaI site at the end of the replaced 1.5 kb portion. The Beta-Geo fusion protein expression is totally dependent on the endogenous promoter. The disrupted gene does not have any detectable protein products. The primary sequence of the mutant allele was depositied in the GenBank database (www.ncbi.nlm.nih.gov/genbank) under accession number HM365221.
