We used the widely used E14 ES cell line for the deletion. The floxed MAZR allele was deleted in ES cells with CMV-Cre (using transient transfection), therefore we generated a germ-line knockout. molecularNote | A floxed neo cassette was inserted within intron 1 and a single loxP site 5' to exon 1 by homologous recombination. Exon 1 and the neo cassette were removed by cre mediated recombination. |
