| molecularNote | Two loxP sites were introduced to flank exon 1 by recombineering. An engineered FRT-Neo-FRT selection cassette was immediately preceding the second loxP site. The first exon was deleted by Cre-mediated recombination. RT-PCR and immunostaining confirmed the absence of mRNA and protein expression in the testis of homozygous mice. Western blots performed on lysates of mouse lung, kidney, heart, spleen, and testis confirmed the loss of the protein expression. |
