molecularNote | Exon 5, coding for part of the Ig-alpha cytoplasmic domain, was targeted with T>G and AC>GT nucleotide substitutions resulting in the amino acid substitutions of serine with alanine at positions 191 and 197 (p.S191A, p.S197A) and threonine with valine at position 203 (p.T203V), respectively. The targeting vector consisted of the wild-type gene with a loxP site inserted into intron 3, a loxP site flanked neomycin resistance gene cassette downstream of the gene and a copy of the 3' end of the gene starting in intron 3 and including a mutated exon 5. Subsequent cre-mediated recombination deleted the wild-type exons 4 and 5 and the neo cassette, leaving a complete gene coding for the mutated peptide. |
