A loxP site was inserted upstream of exon 4. An FRT-flanked neo cassette with a 3' loxP site was inserted downstream of exon 4. Cre-mediated recombination removed exon 4 and the neo cassette. RT-PCR confirmed the absence of transcript expression in the heart.
