molecularNote | This transgenic insertion comprises 10 copies, as determined by dot blot and Southern blot analysis, of a 3.7-kb construct containing the mouse pre-B lymphocyte gene 1 (Vprb1) promoter and enhancer (nt -214 to +142, counting the 5'-most transcriptional initiation site as +1) driving expression of the human interleukin 2 receptor, beta (IL2RB, also called CD122) cDNA, which is followed by exon 2, intron 2, exon 3 and the polyadenylation signal of the human beta-globin gene. Semi-quantitative reverse transcription- (RT-) PCR analysis reveals transgene expression at high levels in bone marrow (BM) and thymus and low to very low levels in spleen, brain and muscle; signal is undetectable in liver. Similar analysis of sorted BM cells from mice of this line reveals expression in the B220+Igm- pro-/pre-B cell, but not in the B220+Igm+ immature/mature B cell population, concordant with the expression pattern of native Vpreb1. Flow cytometric analysis demonstrates that the human IL2RB marker is present on ~10% of B220low/int (characteristic of pro-/pre- and immature B) cells, but not on B220high (mature B) cells nor on B220- cells. |
