molecularNote | A targeting vector constructed by BAC recombineering methods was used to swap the Notch2 intracellular domain (N2ICD) with the ICD of Notch1 (N1ICD) encoding amino acids 1,750 to 2,531, by homologous recombination in B6-derived ES cells. The 3' UTR of Notch2 was not switched so that mRNA stability could be retained. After establishment of germline transmission of this allele, the frt-flanked neo marker that had been introduced as part of the targeting vector was removed by crossing mice to flippase deleter mice. |
