molecularNote  The targeting construct includes a loxP site inserted into intron 1 and the following inserted into intron 2: loxP2 - inverted FRT1 - loxP1 - FRT2 - inverted neomycin casstte - inverted FRT1 - inverted Syt9 coding sequence - inverted loxP1 - inverted loxP2 - FRT2. The Syt9 cDNA incorporated in the allele (intended to function as a conditional knock-in) has been found to be non-functional. Owing to the size of the Syt9 cDNA and the possible presence of cryptic splice donor and acceptor sequences, the conditional Syt9 knock-in strategy activated by FRT1 FLP-recombination does not work.
