| molecularNote | A single loxP site was inserted 5' to the single coding exon, and a loxP-flanked neomycin selection cassette was inserted 3' to the coding exon, but 5' to the noncoding 3'-UTR exon. Germline deletion of the coding exon and the neomycin cassette was achieved by mating 3loxP mice with an EIIa-Cre line which expresses Cre recombinase under the control of the adenovirus EIIa promoter in mouse embryos. Immunoblot analysis of testes lysates confirmed the absence of protein expression in homozygous mutant males. Immunohistochemistry revealed that protein is undetectable in retina sections and seminiferous tubules. |
