molecularNote | CRISPR/Cas9 technology was used to target a construct containing the coding sequence of tdTomato, an orange fluorescent protein, downstream of self-cleaving peptide T2A to exon 1 of Ifnl3. Inadvertently, a 7.2-kb deletion was introduced 5' of Ifnl3, resulting in removal of exon 1 of Ifnl2. Ifnl2 and Ifnl3 are adjacent genes in an inverted orientation, with the genes and flanking genomic regions sharing high sequence identity. Due to the sequence similarity, the gRNAs used disrupted the Ifnl3 locus with insertion of the reporter gene and also unintentionally caused a large deletion in Ifnl2. The reporter is not functional. |
