| molecularNote | A loxP site and a PKG-neomycin resistance cassette flanked by FRT sites were inserted in intron 10 and a second loxP site was inserted in intron 11. Cre-mediated recombination removed exon 11 and the selection cassette. Exon 11 was targeted for deletion to eradicate specifically all actin nucleating products of the gene but not the putative truncated form which lacks all actin-binding WH2 domains and would have functions unrelated to actin nucleation. However, no truncated protein is detected in the brain, indicating a knock-out allele. |
