molecularNote | Site-specific PhiC31 integrase-mediated transgenesis is used to insert a 3,225-bp mouse Cdkn1a (p21) promoter fragment followed by a bicistronic message consisting of the coding sequence for Cre fused to an ERT2 domain, an IRES followed by an ORF coding for enhanced GFP into the intergenic site 2 locus (Hipp11). This construct was microinjected into the pronucleus of recipient zygotes containing attP sites in the Hipp11 locus. Site-specific recombination occurred between the attB sites in the plasmid and attP sites in the H11 genomic locus. Positive embryonic stem cell clones were then implanted into C57BL/6 females. |
