| molecularNote | The transgenic construct contains the mouse Foxf1 gene (bacterial artificial chromosome clone, RP23-55G4) modified by BAC recombineering methods to insert the tdTomato coding sequence, a polyA sequence, an FRT-flanked PGK neo:gb2:kanamycin inserted into exon 1 of the Foxf1 gene. Transient flp infection removed the neo and kanamycin cassette. The final engineered BAC used for transgenic mouse production contained approximately 116,000 base pairs of DNA upstream from the tdTomato start site. The construct is followed by a chloramphenicol selection marker. The BAC clone does not include any other known genes. |
