| molecularNote | A loxP site was inserted upstream of exon 10 and a loxP/FRT-flanked neomycin selection cassette was inserted downstream of exon 10. Exon 10 and the neomycin selection cassette were removed via cre-mediated recombination. Exon 10 constitutes part of the putative catalytic domain and excision of exon 10 is predicted to cause a frame-shift mutation that generates a catalytically inactive protein. RT-PCR detected low levels of aberrant transcripts lacking exon 10 and qRT-PCR confirmed low transcripts. Immunoblotting showed the absence of protein in the lungs of homozygotes indicating that this is a null allele. |
