molecularNote  A loxP site was inserted into intron 8 and 3X SV40 polyA signal sequences, an FRT site flanked neomycin resistance gene cassette, a second loxP site, followed by a duplicate of the 3' end of intron 8, cDNA sequence coding for exons 9-11 (up to the internalization motif in the 3' end of exon 11) plus in-frame mCherry reporter cDNA sequence was inserted into exon 12 downstream of the stop codon. The neo cassette was removed through subsequent Flp-mediated recombination. The allele expresses wild-type peptide and only after Cre-mediated deletion of wild-type exons 9-12 will it express the mutated peptide where the sequence from the internalization motif onward has been replace with the mCherry peptide.
