This cosmid clone hybridised weakly to the cDNA probe after washing at low stringency (data not shown) and was examined further on the basis of its putative homology to dystrophin sequences. Genetic analysis of D4Smh6b through a panel of interspecific backcross progeny mice segregating for the X chromosome mutation mdx2 indicated an autosomal pattern of inheritance. We present here the segregation analysis of D4Smh6b in a panel of 98 Mus spretus/ C57BL/6J interspecific backcross mice typed for 20 mouse chromosome 4 loci3 and demonstrate that D4Smh6b maps to the distal end of chromosome 4. Materials and Methods The cosmid library, dystrophin cDNA probe and screening procedures utilised are as described in Ref. 1. Generation of interspecific backcross progeny mice, origins of the chromosome 4 markers and restriction/hybridisation methods are as described in Ref. 3. Results A 2.5kb EcoRI subclone from the D4Smh6b cosmid was hybridised to TaqI-digested DNA samples from 98 (C57BL/6J x Mus spretus) x CS7BL/6J backcross progeny mice previously characterised for 20 markers mapping to MMU4(3). The 2.5kb single-copy fragment detects a 3.5kb C57BL/6J TaqI restriction fragment length variant and 3.0kb Mus spretus TaqI variant on genomic digest filters (data not shown). Fig. 1 summarises all of the haplotype data for the segregation of this restriction fragment length variant in the 98 backcross progeny. Discussion The haplotype analysis presented in Fig. 1 indicates that D4Smh6b maps 1.03 +/- 1.03cM (1 recombinant in 97 chromsomes) distal to Pnd. Another marker, Skv, has been reported to map distal to Pnd(4) but the relationship between Skv and D4Smh6b has not been determined. It has also been reported that in situ hybridization experiments indicate that D4Smh6b hybridises quite close to the telomere of chromosome 4(4). D4Smh6b is one of the most distal markers on mouse chromosome 4. In addition, 4 different microsatellite sequences have been characterised from this cosmid(5) making D4Smh6b a particularly useful marker for the joint physical and genetic mapping of the distal end of mouse chromosome 4.
