Introduction Intracisternal A-particle (IAP) proviral elements (1) are moderately reiterated and widely distributed in the mouse genome. Distinctive IAP element subfamilies, designated lymphocyte-specific or LS elements, are transcriptionally active in normal mouse B- and T-cells (2). Oligonucleotide probes based on sequences within the long terminal repeats of these IAP element subclasses (LS1, LS2, LS3) detect a restricted number of IAP proviral copies in mouse genomic DNA (3). HindIII fragments representing these proviruses have characteristic strain distribution patterns (SDPs) that are polymorphic among different inbred mouse strains, making them useful for multilocus genomic mapping (3). By comparison of their SDPs with those of known genetic markers in RI strains of mice, the IAP proviruses detected by the LS probes, designated Iapls, have been mapped in the BXD (C57BL/6J X DBA2J) (4), CXB (BALBcBy X C57BL/6By) (4), and AXB (A/J X C57BL/6J) BXA (C57BL/6J X A/J) (5) RI strains. Iapls proviruses were mapped in these RI strains to all mouse autosomes and the X Chromosome. Mapping of the Iapls proviruses in the BXH (C57BL/6J X C3H/HeJ) RI set is presented. Materials and methods DNAs from the BXH RI set and progenitor strains were purchased from The Jackson Laboratory DNA Resource, Bar Harbor, ME, USA. Sequences ofthe LS oligonucleotide probes have been published (2). Restriction digestion of DNAs, agarose gel electrophoresis, and hybridization of the oligonucleotide probes to dried gels were done as previously described (4). Data were analyzed using the computer program Map Manager (6). Locus order was determined by minimizing the number of double crossovers, based on previously mapped markers. Results and Discussion HindIII restriction fragment patterns derived from 5' proviral junctions with flanking DNA were used to map individual IAP proviruses. The proviruses represented by the fragments have been designated Iapls, followed by a number indicating the oligonucleotide probe, a hyphen, and then the HindIII fragment number. The restriction fragment patterns detected in the progenitor strain B and H DNAs are shown in Fig. 1, and fragment numbers for mapped loci are shown beside the strain in which they were present. Forty-six Iapls loci were mapped in the BXH RI set. SDPs are shown in Table 1. Of the 26 B strain Iapls loci mapped in the BXH set all but two (Iapls 1-28 and Iaplsl-69) had been previously mapped in other RI sets or in an interspecific backcross (7). Mapping of the B Strain Iapls loci in the BXH set was consistent with results of previous mapping except for Iapls3-8. This provirus had been mapped to Chr 19 in the BXD RI set (4), but could not be positioned on Chr 19 in the BXH RI set, for which the number of previously mapped loci is quite small. However, the Iapls3-8 provirus could also not be assigned to Chr 19 in an interspecific backcross (7). Previous assignment of Iaplsl-28 to Chr 3 using single chromosome minicells (7) has been confirmed by cosegregation of this provirus with a Chr 3 marker in the BXH set. The nearest linked marker for six of the Iapls proviruses was another IAP locus; 2 of these were other Iapls loci and 4 were anonymous IAP loci (Iap5rc5, Iap5rc2, Iap3rc7, and Iap3ra6) mapped by Kaushik and Stoye (8). The latter were defined by polymorphic PCR fragments generated with a primer from a highly conserved region of IAP proviruses and primers from short repetitive sequences in the flanking DNAs. Mapping of Iapls loci in the BXH RI set provides a total of 46 new markers, 26 in the B strain and 20 in the H strain. Figure 1. (Legend).
