Genes for the hormones somatostatin (SST), calcitonin(CT), thyrotropin alpha subunit (TSH-alpha), and luteinizing hormone beta subunit (LH-beta) have been mapped to mouse chromosomes using mouse- Chinese hamster somatic cell hybrids. DNA was isolated from a panel of 15 mouse-Chinese hamster hybrids, cleaved with a specific restriction endonuclease, and probed with either mouse (TSH-alpha) and rat (CT and LH-beta) cDNA clones or a human genomic clone (SST). The 7.8 kb mouse-HindIII fragment detected by the calcitonin cDNA clone segregated concordantly with mouse chromosome 7. The human somatostatin probe detected a 20 kb fragment in EcoRI cleaved mouse DNA that segregated with chromosome 16 in the hybrid cells. TSH-alpha cDNA hybridized to a single 13.4 kb EcoRI fragment in mouse DNA which in the hybrids co-segregated with chromosome 4. The beta subunit gene of luteinizing hormone was detected a s a 1.5 kb EcoRI fragment which segregated concordantly with mouse chromosome 7. Thus, we have mapped the calcitonin gene to chromosome 7, somatostatin gene (SST) to 16, the thyrotropin alpha subunit (TSH-alpha) to 4, and the luteinizing hormone beta subunit gene (LH-beta) to 7. LH-beta appears as though it may lie in a conserved region since chromosome 7 also has genes for glucose phosphate isomerase and peptidase-4 (D) whose homologous genes as well as the LH-beta gene are on human chromosome 19.
