abstractText  Loren C. Skow Full text of MNL contribution: Alpha A crystallin RFLP's in t-haplotypes. In collaboration with Drs. Dorothea Bennett (UT-Austin) and Joe Nadeau (The Jackson Laboratory), we have examined DNA from 17 different t-haplotypes to search for RFLP's detected with the alpha A crystallin cDNA probe (gene symbol, Crya-l)*(Skow & Donner, Genetics 110:723-732). In contrast to most other gene markers in t-haplotypes, the Crya-1 sequences are polymorphic among t-haplotypes. Three classes of restriction patterns were identified using 8 different restriction enzymes and Fech restriction enzyme produced at least two restriction fragment patterns. The most commonly observed class (t12, tw32, tWPA, tw2, tw8, twl, tw7l, tw73, tw75), designated Crya-1t, has not yet been observed among wild-type mice and may represent sequences of the ancestral t chromosome. The next most abundant class (to, t6, tLL1B-l, tw12, tw12tf, tw5, tw120) had restriction patterns like several inbreds (Crya-la ) while a third class, consisting of a single haplotype, tw121, showed patterns indistinguishable from Crya-lb . These results reveal t-haplotypes to be polymorphic at regions other than the lethal genes and suggest that the variation may result from the acquisition of wild-type DNA. *The gene symbol Crya-1 (originally Acry-1) has been changed to conform to the recently published nomenclature for the homologous gene in humans (Quax-Jenken, et al., 1985, Cyt.
