abstractText  Full text of MNL contribution: Of Four Loci Spanning Mouse Chromosome 17, Two Map to the t Complex. We have looked for variation in DNA sequence as reflected by differences in the lengths of restriction fragments which hybridize to a specific probe as a means of generating new genetic markers. Many of these restriction fragment length polymorphisms (RFLPs) have been mapped with appropriate recombinant inbred (RI) lines. We report here the mapping of four RFLPs to unique loci on chromosome 17, two of which are situated within the t complex. The accompanying table shows the RI strain distribution data for these loci. Polymorphic fragments which hybridize to pMK174, a cDNA clone from a testosterone-induced mouse kidney library, define a locus, RP-17,that maps near qk, within the proximal portion of the t complex. Two other polymorphic fragments which hybridize to pMK174 are specific for mice carrying the t complex. A set of polymorphic fragments which hybridize to the genomic clone alpha-globin4 identified the alpha-globin pseudogene locus, Hba-ps4, and positions it within the t complex, near tf, 4 cM distal to RP-17. RP-11, defined by polymorphic fragments which hybridize to a second mouse kidney cDNA clone, pMK1109, maps 4 cM distal to H-2. A possible phosphoglycerate kinase (PGK) pseudogene locus, RP-10, revealed by polymorphic fragments hybridizing to pHPGK-7e, maps 8 cM distal to RP-11. Together these four loci span the region of known polymorphic markers on chromosome 17. In order from the centromere they are: RP-17 (pMK174), Hba-ps4 (alpha-globin4), (pMK1109), and RP-10 (pHPGK-7e). RP-17 and RP-10 represent, respectively, the most proximal and the most distal loci currently available on chromosome 17 in the RI strains. Significantly, the mapping of pMK174 gives us access to the first known transcriptionally active locus in the t complex, apart from the H-2 genes. (E. Mann, R.W. Elliott and C.
