The BXD recombinant inbred (RI) series, one of the largest RI series, has been valuable for detecting and mapping mendelian genes and will be increasingly valuable in the analysis of the quantitative traits and quantitative trait loci (1). Here we report two new RFLP marker SDPs for the BXD series. MATERIALS AND METHODS Genomic DNA was isolated from fresh livers by a standard phenol extraction procedure. Overnight digestion with restriction enzymes followed the manufacturers' recommendations. Two-fold excess of enzyme was added to avoid partial digestion. Southern blotting was performed using Posiblot (Stratagene) and hybridization with radiolabeled probes was performed in glass jars in hybridization incubator (Robins Scientific). DESCRIPTION OF PROBES AND POLYMORPHISMS Prm-1 locus (Chr 16). Plasmid mP1 (mouse protamine-1) was obtained from Dr. N.B. Hecht (2). After digestion with TaqI a band was observed at 3.7 kb for allele B, and 1.1 kb for allele D. No polymorphism was detected with Hind III, Hinfl, MspI or RsaI. DXWasl7 locus (Chr X). Probe 17-A, an anonymous sequence from the mouse library enriched in X-chromosomal sequences by flow sorting, was obtained from Dr. C. Disteche (3). An EcoRI polymorphism was detected, a characteristic band at 5.0 kb for allele B or 2.8 kb and 2.2 kb (faint) bands for allele D. Polymorphisms were detected also with BamHI, Mspl, PstI, PvuII and BglII. However, since the gene has been placed in the proximal region of Chr 16, one would not expect to see linkage with the known Chr 16 markers because they are more distal. The lowest discordance with other SDPs on Chr 16 is 10/26. The typing of this locus extends the regions swept in the BXD RIs. The DXWas17 SDP shows 3/25 recombinants with the X-1inked marker DXNds3 (4). This is consistent with the general placement of this marker.
