abstractText | Full text of Mouse Genome contribution: 8. Linkage of fat to esterase-1. The mutation fat arose in strain HRS/J and was transferred onto the C57BL/Ks background by five cross-intercross cycles of breeding followed by inbreeding at N5 (Coleman and Eicher, manuscript in preparation). Inbreeding is now at F8 and the stock is maintained by heterozygote matings since fat/fat mice are infertile. An analysis of genetic markers which differ between HRS/J and C57BL/Ks showed that fat/fat mice had the genetic markers typical of C57BL/Ks except for Es-1b, which was from HRS. The possible linkage of Es-1b to fat was tested in the colony of mice of the F6, F7, and F8 generations. Among these mice were 20 fat/fat and 30 fat/+ heterozygotes as determined by the production of fat/fat progeny. Each of these mice provided 2 chromosome tests for the linkage of Es-1b and fat. Also tested were 109 mice who were lean and who could have been either +/+ or fat/+. Each of these mice provided only 1 chromosome test. In total, there were 9 crossovers between Es-lb and fat among 209 chromosome tests providing a map distance of 4.3 +/- 1.4. (B. Paigen, D. Coleman). |
