| abstractText | Full text of Abstract. 503. Insulitis-Dependent Redistribution of Heat-Shock Protein, HSP60 inside Beta Cells of NOD mice is associated with the Induction of HSP60 Autoantibodies. KATRINA BRUDZYNSKI, London, Ont. We have recently reported that sera of NOD mice contain autoantibodies to the endogenous HSP60. Under physiological conditions, the HSP60 antigen is located to mitochondria and secretory granules of beta cells but, in islets affected by insulitis, it accumulates rapidly in cytoplasm and on beta cell surface membrane. Using immunogold-electron microscopy we quantitated changes in cytoplasmic HSP60 with progression of insulitis. Mean concentration of cytoplasmic HSP60 in 4, 7 and 13 week old NOD mice representing 0, 85 and 100% incidence of insulitis, increases from 4.6 to 11.2 to 35.9 immunogold grains/um2, respectively (p<0.001). Simultaneously, the time course of an appearance of HSP60 antibodies (Ab) was established by testing 32 sera from 4 to 25week old NOD mice for their cross-reactivity to human recombinant HSP60 on Western blots. HSP60Abs developed in both sexes between 7 and 13 weeks of age, but were found with the highest frequency in sera of 10-15 week-old mice. They declined rapidly with the onset of diabetes. HSP60 Abs were not observed in sera of 4 week-old mice devoid of insulitis nor in 7 week-old males with delayed onset of insulitis. The cytoplasmic HSP60 levels in these cases were as in control mice. We conclude that the induction of the HSP60 Ab is temporarily associated with the insulitis-related redistribution of the HSP60 inside the beta cells, and that the frequency of detection of HSP60 Abs depends on the duration of insulitis in NOD mice. |
