abstractText  The RPCI-23 bacterial artificial chromosome (BAC) library was screened for the presence of the mouse orthologs of genes on human Chromosome 21. BAC clones were selected and verified by PCR analysis. The cytogenetic locations of the mouse genes were determined by FISH mapping of BAC clones containing these genes. An average of twelve metaphase spreads were analyzed to identify chromosomes exhibiting a fluorescent signal. The location of the signal on the chromosome was also checked for consistency. The chromosomes were counterstained with DAPI; the bright fluorescent bands correspond to the dark Giemsa bands. A pseudo G-band image was produced by converting the DAPI image to a black and white image followed by inversion. The target chromosomes were identified by examining these pseudo G-banded chromosomes.
