abstractText  Full text of Abstract. Lyb 6, A NEW MURINE B-CELL ALLOANTIGEN: BIOCHEMICAL CHARACTERISTICS AND DISTRIBUTION. S.W. Kessler,* A. Ahmed and I. Scher, Naval Med. Res. Inst., Bethesda, Md. 20014 We have demonstrated a new Lyb surface antigen, tentatively called Lyb 6, which is detected by antisera produced in CBA/N mice after hyperimmunization with CBA/J spleen cells. Lyb 6 is a glycoprotein that has been identified by its mobility (which is identical to H-2 alloantigens, mol. wt. 45,000, after complete reduction) in SDS-polyacrylamide gel electrophoresis of immune precipitates from radioiodinated cell lysates. Its distribution among more than 20 different strains distinguishes it from other Lyb antigens and shows no correlation with immunoglobulin (Ig) or H-2 allotypes. Furthermore, it is not associated with Beta2 microglobulin, and it is not present on spleen cells of H-2 congenic B10.Br mice. Lyb 6 is found on spleen, Peyer's patches, and lymph node cells, but not thymus, cortisone-resistant thymus, or bone marrow cells. It is defined as an Lyb antigen since it is found on Ig+ but not Ig- adult spleen cells, and it is not detected on spleen cells which survive treatment with anti-Lyb 4 + C (non-B). The recovery of Lyb 6 is reduced approximately by half after pretreatment with anti-Lyb 5 + C, which under optimal conditions causes a similar decrease in the frequency of surviving B cells. This suggests that Lyb 6 is present on most or all adult spleen cells. The absence of Lyb 6 on bone marrow cells and the kinetics of its appearance on spleen cells of developing neonatal mice suggest that this antigen is expressed late in the maturation of B cells.
