abstractText  Full text of Abstract: Recombination mapping on the short arm of the mouse Y chromosome. S.H. Laval, P.H. Glenister, C.E. Thornton and B.M. Cattanach. Genetics Division, MRC Radiobiology Unit, Chilton, Didcot, Oxon, OX11 ORD, UK The mouse mutation Sxra (1) consists of a duplication of the mouse sex determining region which has been transposed to the pseudoautosomal region of the Y. Following meiotic recombination Sxra can cross over to the X chromosome. This results in XX sex reversal (XIXSxra males). Using the T16H translocation (T(X;16)16H) XY mice carrying Sxra on their X chromosome (XSxralY) can be generated. Xsxra1Y mice frequently show illegitimate pairing between the Sxra region on the X chromosome and the short arm of the Y chromosome at male meiosis (2). Here we demonstrate genetically that recombination events can occur between Sxra and the Y short arm and these can result in deletions of Y chromosome short arm material. XY and XXY female mice carrying these deleted Y chromosomes have been produced. We also observe a variable frequency of hermaphroditism with different deleted Y chromosomes. The only apparent change in Y chromosome content is the loss of a number of copies of the repetitive element Sxl. The partial to complete loss of male determining function is hypothesized to be due to the deletion of Sxl repeats bringing Sry into closer proximity with the Y centromere (3). Under this hypothesis Sry would lie between the mouse Y chromosome centromere and the (Zfyl-Ubelyl-Spy-Sfy2) block of genes on the mouse Y chromosome short arm and be separated from the centromere by the bulk of the Sxl repeat units. This provides a system for the mapping of the mouse Y chromosome by recombination and the analysis of Sry expression with respect to sex determination.
